The British No 1 pulled out of last week's Rome Masters at the start of the third set of his second round match with Spain's Marcel Granollers with a lower back problem.
Murray went to visit with specialists on Monday and Tuesday to see what the full extent of the injury was and if he would be fit enough to play in the second grand slam of the year in Paris.
The French Open starts at Roland Garros on Sunday, with the grass court season and Wimbledon hot on its heels, and Murray has decided that it is too much of a risk to give it a go on the clay.
Instead, Murray will rest and recover instead of fighting it out at the French Open, and focus all his attentions on a return for the grass court season and Wimbledon in particular.
"After consultation with his team and medical specialists, Andy has reluctantly withdrawn from the French Open and will instead focus on preparation for the grass court season," a statement read.
World No 2 Murray himself added: "It's a really tough decision and I love playing in Paris, but after seeking medical advice, I am not fit to compete.
"Apologies to the organisers and thanks to everyone for the messages of support. Now my complete focus is on getting back on the court as soon as possible."
